Product introduction
Ribonuclease A is an endoribonuclease that specifically attacks the 3' end of pyrimidine residues on RNA and clears phosphodiester bonds formed with neighboring nucleotides. The end products of the reaction are pyrimidine 3' phosphate and an oligonucleotide with a terminal pyrimidine 3' phosphate. "In the absence of cofactors and divalent cations, rnase A action can be inhibited by placental ribonuclease inhibitors (B1ackburn et al. 1977) or oxovadium-ribonucleoside complexes (Puskas et al. 1982)."
Scope of application
The structure of nucleic acid was determined. RNA sequence analysis; Hydrolyze RNA in protein samples; Purified DNA.
Usage method
The RNase A powder was dissolved in 0.01M NaAc PH5.2 to a final concentration of 10mg/mL, heated in a boiling water bath at 100 ° C for 15min, and slowly cooled at room temperature. The PH was adjusted to about 7.4 with 0.1 volume of 1M Tris-HCl and stored at -20 ° C in aliquots. RNase A precipitates when the concentrated RNase A solution is heated to 100 ° C at neutral PH.
